Background: | TNF alpha is a protein that is released by activated macrophages in response to lipopolysaccharides and has been shown to induce tumor necrosis when administered to mice with tumors. TNF alpha is implicated in the development of septic shock and tissue damage associated with endotoxemia. Under certain conditions, TNF alpha can exist as a multi-unit protein but typically appears as a single 17 kDa band when analyzed using SDS PAGE without reducing agents. TNF alpha shares similarities with Tumor Necrosis Factor beta (lymphotoxin), as both proteins bind to the same receptors and elicit similar cellular responses. TNF alpha demonstrates cytotoxicity towards specific cancer cells, particularly in synergy with interferon-gamma. While the impact of TNF alpha on healthy human cells is limited, it has been shown to directly harm vascular endothelial cells. Additionally, TNF alpha is known to promote the growth of fibroblasts and various cell lines, activate certain immune cells, and induce the production of inflammatory mediators such as interleukin 1, prostaglandin E2, and collagenase. |
