Background: |
TNF alpha is a protein released by macrophages stimulated with lipopolysaccharides. When injected into mice with tumors, it induces tumor necrosis. TNF alpha is thought to play a role in the development of pathogenic shock and tissue damage associated with endotoxemia. It typically appears in SDS PAGE as a single 17 kDa band, existing as a multimer of two, three, or five non-covalently linked units. TNF alpha shares receptors and cellular effects with Tumor Necrosis Factor beta (lymphotoxin), a 25 kDa protein. TNF alpha causes cell death in certain transformed cells and works synergistically with interferon-gamma to enhance its cytotoxic effects. While it doesn't affect many normal human cells in culture, it directly harms vascular endothelial cells. TNF alpha also promotes the growth of human fibroblasts and other cell lines, activates certain immune cells and bone cells, and triggers the production of interleukin 1, prostaglandin E2, and collagenase. |
