Background: This antibody inhibits the cytotoxic effects of TNF alpha on L929 cells and demonstrates efficacy in inhibiting tumor growth in mice. It protects against the toxicity induced by TNF alpha. TNF alpha, secreted by lipopolysaccharide-stimulated macrophages, induces tumor necrosis in mice with tumors. It is implicated in pathogenic shock and tissue damage associated with endotoxemia. TNF alpha can exist as multimers and shares receptors with TNF beta. It is cytolytic to certain transformed cells and works synergistically with interferon gamma. While it may not affect normal human cells significantly, it can be toxic to vascular endothelial cells. TNF alpha can also stimulate cell growth, activate certain immune cells, and induce the production of various biochemicals like interleukin 1, prostaglandin E2, and collagenase.
