Background: TNF alpha, produced by macrophages activated by lipopolysaccharides, induces tumor necrosis upon injection into tumor-bearing mice. Structurally, TNF alpha forms multimers of two, three, or five units, yet appears as a single 17 kDa band on SDS PAGE under non-reducing conditions. It triggers cell death in specific transformed cells and exhibits direct toxicity towards vascular endothelial cells while having minimal impact on many normal human cells in culture. Additionally, TNF alpha promotes the growth of human fibroblasts and other cell types, activates immune cells like neutrophils and osteoclasts, and stimulates the production of inflammatory mediators such as interleukin 1, prostaglandin E2, and collagenase.
