Purity: > 98% by SDS-PAGE and HPLC analyses.
Biological Activity: Assay #1: Fully biologically active when compared to standard. The ED50 as determined by the dose-dependent stimulation of the proliferation of murine NFS-60 cells is less than 5.0 ng/mL, corresponding to a specific activity of > 2 × 105 IU/mg.Assay #2: The ED50 as determined by a cell proliferation assay using human TF-1 cells is less than 0.05 ng/mL, corresponding to a specific activity of > 2.0 × 107 IU/mg.
Physical Appearance: Sterile filtered white lyophilized (freeze-dried) powder.
Formulation: Lyophilized from a 0.2 µm filtered concentrated solution in PBS, pH7.4, 5% trehalose.
Endotoxin: Less than 1 EU/µg of rRhIL-3 as determined by LAL method.
Reconstitution: Centrifuge the vial briefly before opening to ensure that the contents settle at the bottom. Reconstitute in sterile distilled water or aqueous buffer containing 0.1% BSA to a concentration of 0.1-1.0 mg/mL. Divide the resulting stock solution into working aliquots and store them at or below -20°C. For further dilutions, use appropriate buffered solutions.
Stability & Storage: Use a manual defrost freezer and avoid repeated freeze-thaw cycles.
12 months from date of receipt, -20 to -70°C as supplied.
1 month, 2 to 8°C under sterile conditions after reconstitution.
3 months, -20 to -70°C under sterile conditions after reconstitution.
Synonyms: Hematopoietic Growth Factor, MCGF, Multipotential Colony-stimulating Factor, P-cell-stimulating Factor
Background: Interleukin-3 (IL-3) is a cytokine, a biological signal, encoded by the IL-3 gene found on chromosome 5. It's primarily produced by activated T-cells, human thymic epithelial cells, activated murine mast cells, murine keratinocytes, and neurons/astrocytes. This protein regulates the production, differentiation, and function of two related white blood cell populations – the granulocytes and the monocytes-macrophages. The rhesus macaque IL-3, commonly known as a monomer, consists of 124 amino acids within a single non-glycosylated polypeptide. It's important to note that recombinant rhesus macaque, human and murine IL-3 share minimal homology.
