Purity: > 95% by SDS-PAGE and HPLC analyses.
Biological Activity: Fully biologically active when compared to standard. The ED50 as determined by a cell proliferation assay using murine D10S cells is less than 0.5 ng/mL, corresponding to a specific activity of > 2.0 × 106 IU/mg.
Physical Appearance: Sterile filtered white lyophilized (freeze-dried) powder.
Formulation: Lyophilized from a 0.2 µm filtered concentrated solution in 20 mM Tris, 300 mM NaCl, pH8.5.
Endotoxin: Less than 1 EU/µg of rRtIL-33 as determined by LAL method.
Reconstitution: Centrifuge the vial briefly before opening to ensure that the contents settle at the bottom. Reconstitute the vial with sterile distilled water or an aqueous buffer containing 0.1% BSA to achieve a concentration of 0.1-1.0 mg/mL. Divide the resulting stock solution into working aliquots and store them at or below -20°C. For further dilutions, use appropriate buffered solutions.
Stability & Storage: Use a manual defrost freezer and avoid repeated freeze-thaw cycles.

Background: Part of the IL-1 superfamily, IL-33, also known as NF-HEV and DVS 27, is a proinflammatory cytokine that may influence gene transcription and stimulates type 2 cytokine production from helper T cells, mast cells, eosinophils, and basophils. IL-33’s in vivo administration is linked to severe changes in mucosal organs, with constitutive expression in smooth muscle and airway epithelia and binding to the ST2 receptor. Recombinant rat IL-33, with 156 amino acids, shares 59% and 90% sequence identity with human and murine IL-33, respectively.
