Background: | Sonic Hedgehog (SHH) belongs to the Hedgehog (Hh) protein family, which also includes Desert Hedgehog (DHH) and Indian Hedgehog (IHH). These proteins share a high sequence similarity; for instance, SHH and IHH are 93% identical. SHH expression occurs primarily in the fetal intestine, liver, lung, and kidney, rather than in adult tissues. Structurally, the protein consists of 462 amino acids, including a 23-amino acid signal peptide at the N-terminus. It undergoes cleavage to form SHH N-Terminus and C-Terminus fragments. SHH plays crucial roles in development, functioning as a morphogen that regulates the patterning of various systems such as the limb, midline structures of the brain and spinal cord, the thalamus via the zona limitans intrathalamica, and tooth development. In the absence of Sonic Hedgehog, the patched receptor suppresses the constitutive signaling activity of smoothened. SHH-N, which retains all known signaling capabilities, can undergo lipid modification without losing its receptor affinity, resulting in a more potent form than the unmodified protein. Recombinant human SHH (rHuSHH) substitutes the natural chemically modified Cysteine residue with an N-terminal Ile-Val-Ile sequence. |
