Molecular Weight: Approximately 20.8 kDa, a single non-glycosylated polypeptide chain containing 183 amino acids.
Purity: > 98% by SDS-PAGE and HPLC analyses.
Biological Activity: Fully biologically active when compared to standard. The ED50 as determined by a cell proliferation assay using TF-1 cells is less than 1.0 ng/mL. The specific activity of recombinant human IL-6 is > 1.0 × 108 IU/mg, which is calibrated against rHuIL-6 WHO International Standard (NIBSC code: 21/308).
Physical Appearance: Sterile filtered white lyophilized (freeze-dried) powder.
Formulation: Lyophilized from a 0.2 µm filtered concentrated solution in 20 mM Tris-HCl, 150 mM NaCl, pH8.0.
Endotoxin: Less than 0.01 EU/µg of rHuIL-6 GMP as determined by LAL method.
Reconstitution: Centrifuge the vial briefly before opening to ensure that the contents settle at the bottom. Reconstitute the vial with sterile distilled water or an aqueous buffer containing 0.1% BSA to achieve a concentration of 0.1-1.0 mg/mL. Divide the resulting stock solution into working aliquots and store them at or below -20°C. For further dilutions, use appropriate buffered solutions.

Background: Interleukin-6 (IL-6), a human interleukin encoded by the IL6 gene, can function as a pro-inflammatory and anti-inflammatory cytokine. This interleukin is secreted by T cells and macrophages to stimulate the body's immune response. Notably, IL-6 has a crucial role in the final differentiation of B-cells into Immunoglobulin (Ig) secreting cells, which are involved in lymphocyte and monocyte differentiation. Moreover, IL-6 promotes the growth of myeloma and plasmacytoma, and induces differentiation in nerve cells, B-cells, T-cells, hepatocytes, hematopoietic progenitor cells, and cells of the central nervous system (CNS). The human IL-6 is a non-glycosylated single polypeptide chain of 183 amino acids that signals through a cell-surface type I cytokine receptor complex, which includes the ligand-binding IL-6Rα chain (CD126), and the signal-transducing component gp130 (CD130). It shares around 40% of its amino acid sequence with mouse and rat IL-6 and is equally active on cells of these species.
