Purity: | > 97% by SDS-PAGE and HPLC analyses. |
Biological Activity: | Fully biologically active when compared to standard. The ED50 as determined by inducing IL-8 secretion in human preadipocytes is less than 10 ng/mL, corresponding to a specific activity of > 1.0 × 105 IU/mg. |
Physical Appearance: | Sterile filtered white lyophilized (freeze-dried) powder. |
Formulation: | Lyophilized from a 0.2 μm filtered concentrated solution in 2 × PBS, pH7.4. |
Endotoxin: | Less than 1 EU/μg of rHuIL-36β, 153a.a. as determined by LAL method. |
Reconstitution: | Centrifuge the vial briefly before opening to ensure that the contents settle at the bottom. Reconstitute the vial with sterile distilled water or an aqueous buffer containing 0.1% BSA to achieve a concentration of 0.1-1.0 mg/mL. Divide the resulting stock solution into working aliquots and store them at or below -20°C. For further dilutions, use appropriate buffered solutions. |
Stability & Storage: | Use a manual defrost freezer and avoid repeated freeze-thaw cycles. 12 months from date of receipt, -20 to -70°C as supplied. 1 month, 2 to 8°C under sterile conditions after reconstitution. 3 months, -20 to -70°C under sterile conditions after reconstitution. |

Background: | Interleukin-36 (IL-36), a pro-inflammatory cytokine, plays a pivotal role in the pathophysiology of various illnesses. IL-36α, IL-36β, and IL-36γ, all IL-1 family cytokines, transmit signals through IL-1Rrp2 and IL-1RAcP receptors. IL-36β is found in higher concentrations in psoriatic plaques compared to unaffected psoriatic skin or normal skin and can induce IL-6 and IL-8 production in synovial fibroblasts, articular chondrocytes, and mature adipocytes. It exists in two forms; IL-36β isoform 2 has a potential N-linked glycosylation site at its C-terminus, whereas isoform 1 lacks this site and four conserved β-strands. Human IL-36β isoform 2 displays 62%, 67%, 63%, and 59% amino acid identity with the corresponding isoforms in mice, dogs, cattle, and horses, respectively. |
