Reconstitution: | We recommend that this vial be briefly centrifuged prior to opening to bring the contents to the bottom. Further dilutions should be made in appropriate buffered solutions. |

Background: | The interleukin-3 receptor (IL-3 R) is a heterodimer consisting of a 70 kDa IL-3 R alpha subunit (IL-3 Rα or CD123) and a 120-140 kDa IL-3 R beta subunit (IL-3 Rβ or CD131). IL-3Rα, a glycoprotein part of the hematopoietic receptor superfamily, binds IL-3 with low affinity. However, in the presence of IL-3 Rβ, IL-3 Rα binds IL-3 with much higher affinity. Recent studies show that CD123, the IL-3 Rα, is highly expressed in leukemic stem cells (LSCs) but not in normal hematopoietic stem cells (HSCs). This expression is linked to treatment response, minimal residual disease (MRD) detection, and prognosis. Additionally, CD123 is a crucial marker for identifying and targeting LSCs in refractory or relapsed leukemia. |
