Biological Activity: | Fully biologically active when compared to standard. Measured by its ability to enhance IFN-gamma secretion in NK-92 human natural killer lymphoma cells. The ED50 for this effect is ≤ 2 ng/mL. The specific activity of recombinant human IL-21 > 5.0 × 105 U/mg. |
Physical Appearance: | Sterile filtered white lyophilized (freeze-dried) powder. |
Formulation: | Lyophilized from a 0.2 µm filtered concentrated solution in PBS, pH7.0. |
Endotoxin: | Less than 0.01 EU/µg of rHuIL-21 GMP as determined by LAL method. |
Reconstitution: | Centrifuge the vial briefly before opening to ensure that the contents settle at the bottom. Reconstitute the vial with sterile distilled water or an aqueous buffer containing 0.1% BSA to achieve a concentration of 0.1-1.0 mg/mL. Divide the resulting stock solution into working aliquots and store them at or below -20°C. For further dilutions, use appropriate buffered solutions. |

Background: | Human interleukin-21 (IL-21), which is encoded by the IL21 gene found on chromosome 4, is a versatile cytokine important in the immune system's response to antigenic stimuli. It is primarily produced by CD4+ T cells and plays a crucial role in regulating different immune cells such as cytotoxic T cells and natural killer cells. Notably, IL-21 can trigger cell division or proliferation in target cells. IL-21 works by binding to the interleukin-21 receptor (IL-21R), which has a gamma chain similar to that in other cytokine receptors like IL-2, IL-4, IL-7, IL-9, and IL-15. The interaction between IL-21 and IL-21R initiates a series of reactions, activating the tyrosine kinases JAK1 and JAK3, which in turn activates the transcription factors STAT1 and STAT3. IL-21 is significantly associated with various clinical conditions, including cancer immunotherapy, viral infections, and allergic reactions. |
