Source: | Escherichia coli |
Molecular Weight: | Approximately 15.4 kDa, a single non-glycosylated polypeptide chain containing 133 amino acids. |
Purity: | > 98% by SDS-PAGE and HPLC analyses. |
Biological Activity: | Fully biologically active when compared to standard. Measured in a cell proliferation assay using CTLL-2 cells. The specific activity of recombinant human IL-2 is > 1.0 × 107 IU/mg, which is calibrated against rHuIL-2 WHO International Standard (NIBSC code: 86/500). |
Physical Appearance: | Sterile filtered white lyophilized (freeze-dried) powder. |
Formulation: | Lyophilized from a 0.2 µm filtered concentrated solution in 20 mM PB, pH3.5. |
Endotoxin: | Less than 0.01 EU/µg of rHuIL-2 GMP as determined by LAL method. |
Reconstitution: | Centrifuge the vial briefly before opening to ensure that the contents settle at the bottom. Reconstitute in sterile distilled water to a concentration of 0.1-1.0 mg/mL. If animal-origin-free condition is expected in your product, then sterile distilled water is recommended. Stock solutions should be apportioned into working aliquots and stored at ≤ -20°C. Divide the resulting stock solution into working aliquots and store them at or below -20°C. For further dilutions, use appropriate buffered solutions. |
Stability & Storage: | Use a manual defrost freezer and avoid repeated freeze-thaw cycles. 12 months from date of receipt, -20 to -70°C as supplied. 1 month, 2 to 8°C under sterile conditions after reconstitution. 3 months, -20 to -70°C under sterile conditions after reconstitution. |
Background: | IL-2 is a potent immunoregulatory lymphokine that is generated by T-cells in response to either antigenic or mitogenic stimuli. It is produced by various cells including CD4+ and CD8+ T cells, γδ T cells, B cells, dendritic cells, and eosinophils. IL-2 and IL-2R interaction is crucial for T-cell proliferation and other fundamental immune response functions. There are three subunits to the IL-2 receptor (IL2Rα 55 kDa, IL2Rβ 75 kDa, γc/IL2Rγ 64 kDa common gamma chain) which exist in varying complexes on the cell surface. Mature human IL-2 shares 56% of its amino acid sequence with mouse IL-2 and 66% with rat IL-2. Notably, human and mouse IL-2 displays cross-species functionality. |
