Neuregulin/Heregulin constitutes a family of closely related polypeptide growth factors derived from alternatively spliced genes (NRG1, NRG2, NRG3, and NRG4). Over 14 soluble and transmembrane proteins have been identified originating from the NRG1 gene. The extracellular domain of transmembrane NRG1 isoforms undergoes proteolytic processing, releasing soluble growth factors. HRG1-β1, for instance, features an Ig domain and an EGF-like domain crucial for binding directly to receptor tyrosine kinases erb3 and erb4. This binding prompts heterodimerization of erb3 and erb4 with erb2, activating intrinsic kinase activity that triggers tyrosine phosphorylation. While the precise biological effects of HRG1-β1 are not fully elucidated, it has been observed to enhance motility and invasiveness in breast cancer cells, potentially involving up-regulation of autocrine motility-promoting factor (AMF) expression and function. Recombinant Human Heregulinβ-1 (HRG1-B1) is a 7.5 kDa polypeptide consisting solely of the EGF domain of heregulinβ-1, spanning 65 amino acid residues. |
