Biological Activity: | Fully biologically active when compared to standard. The ED50 as determined by inhibiting IL-1α-dependent proliferation of murine D10S cells is less than 3.0 μg/mL, corresponding to a specific activity of > 333 IU/mg in the presence of 50 pg/mL rHuIL-1α. |
Physical Appearance: | Sterile filtered white lyophilized (freeze-dried) powder. |
Formulation: | Lyophilized from a 0.2 µm filtered concentrated solution in PBS, pH7.4. |
Endotoxin: | Less than 1 EU/µg of rEqIL-1RA as determined by LAL method. |
Reconstitution: | Centrifuge the vial briefly before opening to ensure that the contents settle at the bottom. Reconstitute the vial with sterile distilled water or an aqueous buffer containing 0.1% BSA to achieve a concentration of 0.1-1.0 mg/mL. Divide the resulting stock solution into working aliquots and store them at or below -20°C. For further dilutions, use appropriate buffered solutions. |
Stability & Storage: | Use a manual defrost freezer and avoid repeated freeze-thaw cycles. 12 months from date of receipt, -20 to -70°C as supplied. 1 month, 2 to 8°C under sterile conditions after reconstitution. 3 months, -20 to -70°C under sterile conditions after reconstitution. |

Background: | IL-1RA, initially termed the IL-1 inhibitor, is a member of the interleukin 1 cytokine family encoded by the IL1RN gene. Secreted by diverse cell types such as immune cells, epithelial cells, and adipocytes, IL-1RA inhibits interleukin-1 activity by binding to the IL1R1 receptor and preventing its association with the IL1RAP coreceptor for signaling. It is also employed in treating rheumatoid arthritis, where IL-1 is a key factor. Equus caballus IL-1RA, a single non-glycosylated polypeptide chain comprising 152 amino acids, blocks IL-1-induced inflammatory responses both in vitro and in vivo. It exhibits 26% amino acid homology with IL-1β, 19% with IL-1α, and shares 78%-80% sequence identity with murine, rat, and porcine IL-1RA. |
