Background: Tumor necrosis factor (TNF), also known as tumor necrosis factor isoform 1, is the prototypical ligand of the TNF superfamily. This pleiotropic molecule is crucial in inflammation, apoptosis, and immune system development. It is produced by various immune and epithelial cells and is assembled intracellularly into a noncovalently linked homotrimer that is expressed on the cell surface. Surface-bound TNF can induce the lysis of neighboring tumor and virus-infected cells and can trigger its own downstream signaling upon binding with soluble TNFR I. Additionally, TNF promotes inflammatory responses by activating vascular endothelial cells and macrophages. Recombinant Mouse TNF, fused to a His-tag at the C-terminus, was expressed in insect cells and purified using conventional chromatography techniques.
