Background: Recombinant tumor necrosis factor α/TNF-α is a cytokine devoid of glycosylation, synthesized using recombinant DNA technology in E. coli. Comprised of three identical polypeptide chains, each containing 158 amino acids, TNF-α forms a compact, bell-shaped homotrimer. The individual subunits possess a relative molecular mass of 17,484 Daltons each. This potent lymphoid factor exhibits cytotoxic effects on a broad spectrum of tumor cells and select target cells. TNF-α, with a subunit molecular mass of 17 kDa, significantly influences growth regulation, differentiation, inflammation, viral replication, tumorigenesis, autoimmune disorders, and responses to viral, bacterial, fungal, and parasitic infections. Beyond inducing hemorrhagic necrosis in tumors, TNF-α is implicated in tumorigenesis, tumor metastasis, viral replication, septic shock, fever, inflammation, and autoimmune conditions like Crohn's disease, rheumatoid arthritis, and graft-versus-host disease.
