Background: | IL-12 (interleukin 12) is a heterodimeric cytokine made up of a 35-kDa light chain (p35 or IL-12A) and a 40-kDa heavy chain (p40 or IL-12B). The p35 gene is located on chromosome 3 in humans and chromosome 6 in mice, while the p40 gene is on chromosome 5 in humans and chromosome 11 in mice. The IL-12 receptor, which binds IL-12, consists of two subunits, IL-12RB1 (CD212) and IL-12RB2. IL-12 is a pro-inflammatory cytokine that promotes interferon-γ (IFN-γ) production, and T helper 1 (TH1) cell differentiation, and links innate and adaptive immunity. It is produced by dendritic cells and phagocytes in response to pathogens. The production of IL-12 is regulated by gene expression mechanisms, Toll-like receptor (TLR) patterns, and interactions between different dendritic cell subsets involving cytokines like IL-10 and type I IFN. |
