Characteristics: The heterodimer human IL-12B & IL-12A is produced by co-expression of IL-12B and IL-12A, has a calculated MW of 36.6 kDa (IL-12B) and 23.8 kDa (IL-12A). Subunit IL-12B is fused with his tag at the C-terminus and subunit IL-12A is fused with flag tag at the C-terminus. The predicted N-terminus is Ile 23 (IL12B) and Arg 23 (IL12A). The reducing (R) heterodimer protein migrates as 33-47 kDa due to glyosylation.

Background: Interleukin 12 (IL-12), also referred to as p70, is a cytokine naturally synthesized by dendritic cells, macrophages, and human B-lymphoblastoid cells (NC-37) in response to antigenic stimulation. It exists as a heterodimeric structure comprising IL-12A (p35) and IL-12B (p40) subunits. IL-12 primarily facilitates the differentiation of naive T cells into Th1 cells and is recognized as a potent T cell-stimulating factor, promoting T cell growth and functionality. Additionally, it induces the production of IFN-γ and TNF-α from T cells and NK cells, while attenuating IL-4-mediated suppression of IFN-γ. IL-12 significantly contributes to the activities of natural killer cells and T lymphocytes. Moreover, it exhibits anti-angiogenic properties, impeding the formation of new blood vessels.
