Source: | Escherichia coli |
Molecular Weight: | Approximately 17.4 kDa, a single non-glycosylated polypeptide chain containing 152 amino acids. |
Purity: | > 98% by SDS-PAGE and HPLC analyses. |
Biological Activity: | Full biological activity when compared with standard protein by stimulating proliferation of PHA-activated human peripheral blood mononuclear cell (PBMC). The specific activity of recombinant human IL-7 is > 1.0 × 108 Units/mg, which is calibrated against rHuIL-7 WHO reference reagent (NIBSC code: 90/530). |
Physical Appearance: | Sterile filtered white lyophilized (freeze-dried) powder. |
Formulation: | Lyophilized from a 0.2 µm filtered concentrated solution in 20 mM PB, 150 mM NaCl, pH7.4. |
Endotoxin: | Less than 0.01 EU/µg of rHuIL-7 GMP as determined by LAL method. |
Reconstitution: | Centrifuge the vial briefly before opening to ensure that the contents settle at the bottom. Reconstitute the vial with sterile distilled water or an aqueous buffer containing 0.1% BSA to achieve a concentration of 0.1-1.0 mg/mL. Divide the resulting stock solution into working aliquots and store them at or below -20°C. For further dilutions, use appropriate buffered solutions. |
Stability & Storage: | Use a manual defrost freezer and avoid repeated freeze-thaw cycles. 12 months from date of receipt, -20 to -70°C as supplied. 1 month, 2 to 8°C under sterile conditions after reconstitution. 3 months, -20 to -70°C under sterile conditions after reconstitution. |
Background: | Interleukin-7 (IL-7), which is produced by the IL7 gene and secreted by stromal cells in the red bone marrow and thymus, binds to the IL-7 receptor. This is a heterodimer composed of IL-7 receptor alpha and IL-2 receptor gamma chain. IL-7 encourages the development of hematopoietic stem cells into lymphoid progenitor cells and also promotes the proliferation of B cells, T cells, and NK cells. Mouse IL-7 has roughly 65% similarity in amino acid sequence with human IL-7, and both proteins show cross-species activity. The potential of IL-7 as a target for immunotherapy has been explored in numerous clinical trials in humans for a variety of cancers and during HIV infection. |
